The purpose of this study is to understand the mechanism behind metabolite synthesis and to analyze the metabolite biosynthesis pathway. Basically the metabolite expressions in the plants are affected by a cascade of pathway in synthesis reactions with various other factors like environmental, expression level of enzymes etc. In this study, we choose Coscinium fenestratum (Gaertn.) Colebr, (daruharidra) as study object and leaf and stem tissues are selected as samples. Coscinium fenestratum (Gaertn.) Colebr, commonly called as daruharidra is considered as a critically endangered/regional medicinal plant belonging to the family Menispermaceae. It is a highly traded medicinal plant and its stem and root is used in more than 60 ayurvedic preparations to cure variety of ailments such as ulcers, skin diseases, eye disorders, inflammation, hypertension, jaundice, diabetes, snake bites etc., and is widely used in Ayurveda, folk, Tibetan and Sidha medicine system. Dried stem, leaves and root form are the useful parts for medicinal purpose due to the presence of Benzylisoquinoline alkaloids (BIAs). We sequenced the stem and leaf transcriptome of C.fenestratum using Illumina HiSeq 2000 paired-end sequencing followed by de novo transcriptome assembly. Multiple databases were used to obtain a comprehensive annotation and the transcripts were functionally classified using GO, KOG and PlantCyc. The metabolite biosynthesis pathway and its mechanism of synthesis is analysed with the information on enzyme sequence alignment, metabolite expression level, SSR sequence within and between the samples is done in the present work.
