In response to carbon source switching from glucose to non-glucose, such as ethanol and galactose, yeast cells can directionally preprogram cellular metabolism to efficiently utilize the nutrients. However, the understanding of cellular responsive network to utilize a non-natural carbon source, such as xylose, is limited due to the incomplete knowledge on the xylose response mechanisms. Here, through engineering the xylose assimilation module along with its match with local metabolism, the generated xylose strain could grow on xylose medium but exhibited extremely low growth rate due to the unoptimized the cellular metabolic network. Through performing adaptive evolution, the evolved populations exhibited significantly improved cell performances on xylose. Whole-genome sequencing of the evolved strains further helps us to understand the reprogramming mechanism on xylose.
