Lymphocytes were isolated from the spleen of mice. CD8+ T cells were purified either by FACS Aria cell sorter (BD Biosciences) or by MACS negative selection EasySep. CD8+ T cells were then cultured in complete RPMI-1640 culture medium with 10 ng/ml recombinant mouse IL-2 in plate pre-coated with anti-CD3 (2 ug/ml) and anti-CD28 (1 ug/ml) for 48 hr. Cells were then washed with PBS and expanded with IL-2 treatment.
