Pseudomonas aerugÂiÂnosa is an opÂporÂtunisÂtic pathogen which infects cysÂtic fiÂbroÂsis and canÂcer paÂtients with comÂproÂmised immune sysÂtems. LasR is a masÂter regÂuÂlaÂtor which conÂtrols the viruÂlence of P. aerugÂiÂnosa in reÂsponse to bacÂteÂrÂial cell-density and host sigÂnals. DurÂing inÂfecÂtion, lasR is frequently muÂtated, conÂferÂring P. aerugÂiÂnosa a growth adÂvanÂtage in hosts and enhances reÂsisÂtance to widely used anÂtibiÂotics. HowÂever, the mechaÂnisÂtic baÂsis of lasR muÂtaÂtion is not well underÂstood. We have tested here the hyÂpothÂeÂsis that transcription strength is a contribÂuÂtory deÂterÂmiÂnant of lasR muÂtaÂgenÂeÂsis. P. aerugÂiÂnosa strains with difÂferÂent lasR tranÂscription strengths were therefore enÂgiÂneered and the lasR muÂtaÂtions were monÂiÂtored unbiasedly usÂing next-genÂerÂaÂtion seÂquencÂing technolÂogy.
