To compare the transcriptomic profile of mouse spinal cord homegenates from different conditions using the NanoString nCounter Mouse Glial Profiling Panel with additional customized probes. In this study, we extracted mRNA from mouse spinal cord homogenates from the following groups: wild-type 1-year-old female (WT-OF), wild-type 1-year-old male (WT-OM), wild-type 2–3-month-old female (WT-YF), wild-type 2–3-month-old male (WT-YM), 1-year-old female 10 days post-microglial Tgfbr2 deletion (KO-OF-D10), 1-year-old female 20 days post-microglial Tgfbr2 deletion (KO-OF-D20), 1-year-old female 30 days post-microglial Tgfbr2 deletion (KO-OF-D30), 1-year-old male 30 days post-microglial Tgfbr2 deletion (KO-OM-D30), 2–3-month-old female 30 days post-microglial Tgfbr2 deletion (KO-YF-D30), and 2–3-month-old male 30 days post-microglial Tgfbr2 deletion (KO-YM-D30). The samples were hybridized to the nCounter Mouse Glial Profiling Panel CodeSet (XT-CSO-M-GLIAL-12, NanoString). This CodeSet includes 757 probes for genes of interest, 13 probes for internal reference genes, and 10 additional customized probes for Smad4, Smad7, Nrros, Tgfbr2, Tgfbr3, Smurf1, Tgfb2, Cxcr4, Adgre4, and Clec4b1. A total of 3–5 mice were included for each group.
