Jacobaea vulgaris HSS cDNA sequences were obtained using specific primers and cloning of the obtained amplicons. Seqeuncing of a number of clones revealed presence of three variants of homospermidine synthases, which were denoted HSS2, HSS3 and HSS4. All three were shown to be bioactive using heterologously expressed protein (in E. coli) and performing in vitro enzymatic activity assays using spermidine and putrescine as substrates. The formation of homospermidine was confirmed by HPLC analysis.
