Atractylodes macrocephala is a traditional Chinese medicine in China. It is widely used in clinic and the market demand is great. The quality difference between A. chinensis(Bunge) Koidz and A. japonica Koidz.ex Kitam is great, which seriously affects its clinical curative effect. Determination of Atractylosin by High-performance liquid chromatography. Transcriptomics coupled with Metabonomics for elucidating differences between the components in A. chinensis(Bunge) Koidz and A. japonica Koidz.ex Kitam. It was found that ACS, ACC, PAL and NOS were the key genes and metabolites related to the synthesis of atractylodes macrocephala. The decrease of salicylic acid content in A. chinensis(Bunge) Koidz may lead to the decrease of its ability to mediate the elicitor of endophytic fungi, resulting in the down-regulation of the expression of TGAL4 transcription factor and the up-regulation of DOGL3 transcription factor in A. chinensis(Bunge) Koidz cells, which may affect the accumulation of Atractylosin. The study above provides a theoretical basis for elucidating the biosynthesis pathway of atractylosin in A. chinensis(Bunge) Koidz. Overall design: Samples of A. chinensis(Bunge) Koidz and A. japonica Koidz.ex Kitam were collected in the Herb Garden of Changchun University of Traditional Chinese Medicine, Nanguan District, Changchun City, Jilin Province, China (latitude 43°49'N, longitude 125°25'E). Plant samples were collected using a five-point sampling method. After the plant samples are collected, the surface soil and dust are washed off with clean water, and the surface liquid of the material is blotted with absorbent paper. It is divided into 50-100 mg sizes, divided into two aliquots, one into sterile, enzyme-free cryovials with screwed caps and labeled. It was quick-frozen in liquid nitrogen, and then quickly moved to -80°C for storage for transcriptome and metabolome sequencing. Another sample was naturally dried to determine the content of the pharmacodynamic ingredient. All experimental samples were identified by Associate Professor Qi Weichen of the School of Pharmacy of Changchun University of Chinese Medicine.
