we reported a high-quality chromosome-level genome assembly and annotation of T. fasciatus by using a combination of PacBio-CCS, Hi-C technology, BGISEQ short-read sequencing, and full-length transcriptome. Comparative genomic analyses were conducted to provide insights into its taxonomic position and genetic characteristics associated with its catadromous and short-life traits. The high-quality assembly and well-annotated genome can provide a comprehensive basis for further researches on the genetic mechanism of adaptation and conservation genetic studies, and facilitate the genetic breeding and germplasm identification of the roughskin sculpin. This assembly and annotation will also further establish the roughskin sculpin as a representative species for genetic and genomic studies into ecology and evolution of freshwater sculpins.
