Liver failure (LF) is a severe clinical syndrome resulting from liver diseases, posing a significant health threat with rising incidence rates. However, reliable biomarkers for predicting liver failure are lacking. Extracellular vesicles (EVs) have recently emerged as crucial mediators of intercellular communication, influencing disease progression in liver failure. Yet, it remains unclear whether plasma EVs from liver failure patients contain specific pathogenic molecules reflecting disease status. In this study, we isolated extrachromosomal circular DNA (eccDNA) from plasma EVs of liver failure patients and healthy individuals, comparing characteristics between liver failure EV-derived eccDNA (LFEVs-eccDNA) and healthy control eccDNA (HCEVs-eccDNA) using high-throughput sequencing.
