Liver Transcriptomics from Male and Female HET3 Mice on Control, Low Amino Acid and Low Isoleucine Diets when started in Young and Middle Aged Mice

Mice were put on either a control, low amino acid or low isoleucine diet at 9 weeks of age or 6 months of age and their liver transcriptomic expression was measured after 14 weeks and 18 months respectively on diet after an overnight fast. This was to discover the impact of sex, protein and isoleucine level on the metabolism of the liver. To compare the metabolic effect of total amino acid and just isoleucine restriction in hetergeneous mice on different sexes and ages, 9-week-old or 6-month-old mice were placed on either a Control, Low Amino Acid or Low Isoleucine diets for 13 weeks. All mice were maintained at a temperature of approximately 22°C. Mice were euthanized by cervical dislocation and livers were harvested and snap frozen in liquid nitrogen following overnight (16-hr) fasting.
