Immunomodulatory agents (IMiDs) are a major class of drugs for treating multiple myeloma (MM); however, acquired resistance to IMiDs remains a significant clinical challenge. To study mechanisms of resistance to IMiDs, we generated lenalidomide-resistant (LenR) KMS-11 and MM1.S multiple myeloma cells by gradual dose escalation of lenalidomide until resistance was achieved and performed various analyses including whole exome sequencing using Illumina NovaSeq 6000 platform with PE150 strategy.
