Disclosed are a method for increasing the content of benzylisoquinoline alkaloids in Macleaya cordata by over-expressing a berberine bridging enzyme and the use thereof. The method comprises: amplifying a full-length encoding sequence of a key gene， namely a berberine bridging enzyme (BBE) participating in the metabolism of alkaloids in Macleaya cordata by means of a PCR technique; successfully integrating the BBE gene in Macleaya cordata into a plant expression vector pCAMBIA2301D by combining with an In-Fusion homologous recombination method and an efficient vector construction technique; and then introducing the expression vector into an explant of Macleaya cordata by means of an agrobacterium-mediated method， so as to increase the content of benzylisoquinoline alkaloids in Macleaya cordata.
