AML is a genetically heterogeneous, aggressive hematological malignancy induced by distinct oncogenic driver mutations. Cancer immunosurveillance is a coordinated response from innate and adaptive immune cells against cancer cells. The effect of specific AML oncogenes on immune activation or suppression has not been investigated. Overall design: Bone marrow from AML patients or healhy controls were obtained and Ficoll gradient seperated to extract mononuclear cells. Cells were sorted for immune cell fraction (CD34-CD45+) and subject to 3' single cell RNA sequencing using 10x Genomics platform.
