We have employed a single nucleus sequencing approach using 10x Genomics snRNA-seq to study the process of skeletal muscle hypertrophy. We examined the plantaris muscle of control adult mice (Ctl) and adult mice after 7 days of induced overload (7OV). This study highlights a new subpopulation of myonuclei in the muscles of control and hypertrophied mice. Overall design: We used 3 mice model : C57bl6N (WT), ROSAmTmG;PAX7CREert2/+ (mTmG) and HSA-Cre-ERT2:RhoAflox/flox (Mut). The plantaris muscle underwent compensatory surgery and were removed 7 days later with the control plantaris muscles. Nuclei were sorted by Fluorescence-activated cell sorting (FACS) with DAPI and analyzed using snRNA-seq.
