RNA-sequencing has been used to obtain transcriptome-wide differences that occur in paraspeckle-deficient, MYCN-amplified/model high-risk neuroblastoma cell lines with an increase in the long non-coding paraspeckle-associated NEAT1_2 RNA, to decipher the role of paraspeckles in this cancer type. Overall design: Methods: KELLY cell lines were treated with custom-designed morpholino oligonucleotide:DNA leash lipoplexes, and cells were lysed. RNA-seq was conducted to show transcriptome wide changes occurring due to increased NEAT1_2 levels and subsequent paraspeckle formation. Our study has shown upregulation of differentiation pathways is associated with increased NEAT1_2 RNA and paraspeckle levels.
