Using the RiboTag approach, we profiled the cell type-specific molecular landscape of the outer hair cells and supporting cells as well as the whole cochlea, both before and after noise trauma (6h and 24h) that induces a permanent hearing loss. 10-week-old PrestinCreERT2;RiboTag;CBA and Sox2CreERT2;RiboTag;CBA mice were subjected to a 105dB SPL noise for 2h. Control animals from both strains were not subjected to noise. For the noise group, mice were euthanized and the cochlear tissue dissected 6 hours or 24 hours following noise exposure. Tissue was homogenized, an aliquot was processed for RNA extraction (input sample representing the whole cochlea) and the rest of the homogenized tissue was processed for RiboTag immunoprecipitation using an HA antibody (IP samples). Both input and IP samples were sequenced.
