This data is related to the development of an electrochemical immunosensor for the S protein of SARS-CoV and SARS-CoV-2 viruses. For that, screen-printed carbon electrodes (SPCEs) were modified by the electrodeposition of gold nanoparticles using different conditions. Specific anti-S protein antibodies were immobilized in the surface of the electrode using EDC and NHS, with the presence of the analyte being assessed by electrochemical impedance spectroscopy. Immunsensors were tested using biological samples, showing great potential to be applied in clinical samples. Furthermore, the devices were able to detect S protein in a rapid (35 min), sensitive and low-cost (< US$ 0.03 per device) manner.
