Induced pluripotent stem cells were obtained from an ALS patient with an SOD1A4V mutation. To determine whether the phenotypes we observed were dependent on the SOD1A4V allele, we used a two-step, zinc-finger nuclease (ZFN)-mediated gene targeting strategy to correct the mutation in iPSC line 39b. Stem cells were sequenced at 30X coverage to examine off target cut sites, altered copy number variation and karyotypic stability of the modified line.
