Glutamate-Treated R28 Cell Model and Compound 35 Intervention: RNA Sequencing Analysis

We used the glutamate excitotoxicity as a cell model of glaucoma. We discovered a new natural compound which was protective for glaucoma model. To further explore the mechanisms of the effective effects, we conducted a RNA high throughput sequencing. The R28 cells were collected 24h after glutamate intervention with or without the treatment of compound 35. Each group contained 4 samples in which each sample contained single cell dish. Total RNA was extracted from cultured cells with RNAiso plus reagent (Takara). Total amounts and integrity of RNA were assessed using the RNA Nano 6000 assay kit of the Bioanalyzer 2100 system (Agilent Technologies, CA, USA). After RNA was converted to cDNA, the samples were sequenced by the Illumina NovaSeq 6000 at Novogene.
