Genome-wide CRISPR screen identified NEK6 as a determinant of sensitivity to CDK4/6 inhibitor in endometrial cancer

We used the genome-wide CRISPR screen to identify genes affecting the sensitivity of CDK4/6 inhibitors in endometrial cancer. Analysis of clinical samples and datasets, and in vitro and invo experiments (including CCK8, apoptosis, western blot, q-PCR and mouse models) were applied to confirm the function of the target gene. The RNA-seq and IP-MS assays were utilized to further verify the downstream pathway.
