In this study, we mutated all residues of Zika virus (ZIKV) Envelope protein domain III and stem-anchor, and transfected the cDNA mutant swarm into C6/36 (mosquito) or Vero (monkey) cells to identify potential host-specific mutations that are beneficial for replication in the respective cell lines.
