Epigenetic mechanisms are dynamic processes that govern gene expression in health and disease. DNA methylation, catalyzed by DNA methyltransferases (DNMTs) such as DNMT1, represents a stable but reversible process in which DNA is chemically modified to facilitate or prevent the binding of transcriptional regulators. Despite being a vital component of transcriptional regulation throughout all stages of life in health and disease, the sequence specificity of DNMT-binding is so far poorly understood. Here, we aimed to identify potential binding sites of DNMT1 as well as the dynamics of this process in immortalized, murine cerebellar granule cells, with and without the presence of a recombinant ephrinA5 protein, a ligand known to play a crucial role in neurodevelopment. Overall design: Chromatin immunoprecipitation sequencing for DNMT1 in immortalized cerebellar granule cells treated with Fc control or recombinant ephrinA5-Fc. Inputs were equimolarly pooled per treatment.
