Here, we developed a new method- Single Cell Structure Probing Of RNA Transcripts scSPORT to simultaneously determine RNA secondary structural signal and gene expression in a single cell. We applied it to study RNA structures in hESCs and different stages of neuronal differentiation such as neuronal precursor cells (NPC, seven days post differentiation), immature neurons (iNeu, eight days post differentiation) and early neurons (NEU). Further more, to validate the influence of PUM2, we overexpressed and knock-down PUM2 in hESC cells then probed the RNA structure. The repository contains the bulk samples of RNA structure probing used as benchmarks.
