5' tRNA-fMet1 half is delivered into cystic fibrosis airway epithelium cells by outer membrane vesicles and regulate gene expression. We used chimeric eCLIP to map transcriptome-wide interactions between 5' tRNA-fMet1 half and target mRNAs mediated by AGO2. Overall design: 5' tRNA-fMet1 half or siRNA negative control (siNC) was transfected into cystic fibrosis airway epithelial cells from one donor to generate two samples. For each sample, an input and an immunoprecipitated sub-sample using an anti-AGO2 antibody were generated for cDNA library preparation. To deeply profile the target repertoire of 5' tRNA-fMet1 mediated by AGO2, a 5' tRNA-fMet1 half specific primer was used for targeted sequencing (targeted chimeric eCLIP).
