This project provides the genome assembly of Prunus cerasus L., common name sour cherry (cultivar Schattenmorelle). Sour cherry is an allotetraploid species originating from a hybridization event between the two species Prunus fruticosa and Prunus avium. Description The assembly 20_WGS_PCE_2.0 is based on 300x ONT R9.4.1 PromethION data, 114x of paired-end Illumina HiSeq 4000TM, and PhaseGenomics Hi-C data. The assembly was performed due to the following steps: initial raw data assembly generation with Minimap2 (v2.16-r922) and assembler Miniasm (v0.2-r137-dirty) followed by Racon (vv1.4.10) and Pilon (v1.22) base-quality improvement with raw ONT and Illumina read data to a primary assembly. Separation of the ancestral genomes was performed by mapping publicly available Prunus avium reads to the primary assembly to select contigs that fit the hypothesis of 1 or more clear coverage peaks on all Prunus avium derived contigs. The remaining contigs were then assigned to Prunus fruticosa. The two subgenomes PCE_Avium and PCE_Fruticosa were purged using the purge_dups (V1.0.1). Chromosome-scale scaffolding was performed by Phase Genomics (Seattle, Washington, USA) with Proximo Hi-C. Pseudochromosome-scale scaffolds were assigend by synteny based on Prunus persica (peach) GCA_000346465.2.
