To elucidate the function of calcineurin in dendritic cells (DCs) downstream of C-type lectin receptor, we established mouse model that has DC-specific calcineurin deficiency. We stimulated the bone marrow-derived DCs from these mice with lipoarabinomannan (LAM), a ligand of Dectin-2, and performed RNA sequencing. From multiple genes that showed altered expression pattern in calcineurin-deficient DCs, we found that Il10 transcription increased in the absence of calcineurin. Our further study elucidated the mechanism by which calcineurin regulates Il10 transcription in LAM activated DCs. Overall design: Bone marrow-derived DCs from wild type and calcineurin-deficient mice were stimulated with lipoarabinomannan (LAM) for 0, 2, 4, 8, or 12 h and used for RNA sequencing.
