The presence of DNA methylation in mammalian mitochondrial DNA (mtDNA) would have significant implications for mtDNA gene expression. Currently, there is no consensus on whether mtDNA methylation exists in mammals as there are conflicting data. We have investigated the levels of mtDNA methylation in Sus scrofa mtDNA using next generation bisulfite sequencing on: control mtDNA samples that have been purposely DNA methylated and or left unmethylated; isolated mtDNA samples from ovary and oocytes; and on total DNA from oocytes.
