N-methyl-D-aspartate receptors (NMDARs) are glutamate-gated cation channels with high calcium permeability. Their activity supports megakaryocytes proliferation and maturation, however their downstream effects on the megakaryocyte transcriptome remain poorly elucidated. We used Clariom S Pico Arrays to profile the transcriptome changes caused by loss of Grin1 in mouse bone marrow megakaryocytes. Overall design: Conditional deletion of Grin1, the gene encoding the obligate NMDAR subunit GluN1, in mouse megakaryocytes was directed by the Pf4 promoter using the Cre-LoxP system. This was achieved by breeding Pf4-Cre with Grin1-loxP/loxP mice. Bone marrow megakaryocytes were isolated by magnetic bead isolation, and total RNA isolated before transcriptome changes were profiled by RNA microarray technology.
