Microbiome-dependent tryptophan metabolism suppresses inflammatory immunity in the pancreatic tumor microenvironment by activation of the aryl hydrocarbon receptor [scRNA-seq]

To analyze transcriptome changes in the tumor microenvironment caused by loss of macrophage AhR we performed scRNA sequencing analysis. Overall design: 14 days after tumor implantation, tumors from three Ahrfl/fl or three LysM-CRE cKO mice were digested, pooled, and stained with DAPI. Live cells were FACS-sorted into buffer (PBS + 2% FBS), washed 2 times with PBS + 0.04% BSA, and then only 10x10^3 cells mixed with 10X Genomics Chromium single-cell RNA master mix.
