Multiplexed Sequencing of Expression Cassette Expressing Human PSAT1 in Yeast. PSAT Variant Calling

Multiplexed sequencing of yeast yPSAT1 expression cassette. yPSAT1 encodes the human PSAT1 protein, using a codon-optimized derivative of the human PSAT1 cDNA sequence. Derivatives of yPSAT1 were introduced into haploid yeast strains so that each encoded a version of human PSAT1 with one amino acid changed. Multiplexed sequencing using the MinION platform was used to determine the altered codon, and associated amino acid change, in each yeast transformant.
