Mus musculus Genome sequencing and assembly

Cancer cells are the primary source inducing T cell senescence (Tsen). We established a Tsen-cell-induction in vitro model and applied mouse adipocyte tissue extracts (MATES) to mimic adipocyte-rich tumor microenvironment. We sorted splenic CD8+T cells of wild-type (WT) C57BL/6 mice. Splenic CD8+T cells were pretreated with ConA for 12h, seeded in a 75cm2 flask (1x106), and cocultured with ID8 cells and MATES for 48h. Then suspended CD8+T cells were isolated using a mouse lymphocyte separation kit (Tianjin Haoyang Huake Biotechnology Co., Ltd.). 1 ml TRIzol (Tiangen Biotech, CO., Ltd.) reagent was added to lyse the cells and the samples were sent to Tiangen Biotech, CO., Ltd. for RNA sequencing and analysis.
