A deficiency of ethanolamine plasmalogens (a subclass of ethanolamine glycerophospholipids) has been demonstrated in Alzheimer disease brain and for this reason, we targeted ethanolamine glycerophospholipids in this study. Our present finding is of a relative lack of ethanolamine plasmalogens associated with apolipoprotein E4 compared to E3, using samples from individuals who have no evidence of Alzheimer disease. This result strengthens the evidence that ethanolamine plasmalogen deficiency in Alzheimer disease brain contributes to pathogenesis rather than merely being a consequence of the disease. The risk associated with apolipoprotein E4 may relate at least in part to its reduced ability to compensate for the brain plasmalogen deficit.
