Expression of COL17A1 mRNA encoding collagen XVII is associated with worse or better patient survival depending on cancer types. However, little is known of the underlying mechanisms of a context-dependent effects of collagen XVII on cancer progression. To test this, a murine pancreatic cancer cell line AK4.4 was transduced with Col17a1-expressing vector (AK4.4-Col17a1) or corresponding control vector (AK4.4-vector). Mouse tumorigenicity assay showed that the overexpression of Col17a1 suppressed the growth of AK4.4-Col17a1 tumor relative to AK4.4-vector tumor in recipient FVB mice. To explore the mechanisms of how Col17a1 suppresses the tumorigenicity of AK4.4, we performed RNA-sequencing analysis of AK4.4-Col17a1 and AK4.4-vector tumors isolated from FVB mice.
