The study investigates the influence of Alzheimer's disease brain-derived tau aggregates on human astrocytes, exploring the relationship between tau molecular diversity and astrocytic response. Astrocytes, the most abundant brain cell type, provide critical support for neurons and regulate synaptic activity. In AD, astrocytes show heterogenous responses to tau pathology, yet their role in the disease's progression remains unclear. Using iPSC-derived astrocytes, the study shows that tau aggregates from postmortem AD brain samples are internalized and processed by astrocytes. Tau internalization is associated with distinct gene expression changes in astrocytes, including dysregulation of genes linked to reactive astrocyte states, autophagy, and other pathways indicated in AD. This research provides novel insights into the complex interaction between tau diversity and astrocytic response.
