Metabolic vulnerabilities of aneuploid mammary epithelial cells

We performed genome-wide CRISPR knockout screens in isogenic aneuploid/diploid human mammary epithelial cells to identify synthetic lethalities associated with supernumerary chromosomes. We identify several aneuploidy-associated metabolic vulnerabilities from these CRISPR screens, and perform follow-up studies including RNA-seq analysis to identify stress signaling pathways associated with metabolic stress.
