Fibrinogen deposition promotes metastasis in lung because fibrinogen binds circulating tumor cells and the fibrinogen-tumor cell complex is allowed to invade in the lung tissue. We used microarrays to compare gene expression profiles of the tissues near and apart from fibrinogen deposition in non-cancer patients and cancer patients. Overall design: Formalin fixed paraffin embedded (FFPE) lung tissues from non-cancer and cancer patients. Slide sections were prepared and blood vessels with or without fibrinogen deposition were isolated by using laser microdissection. Total 20,000 vessels were obtained for each group [Fibrinogen(-)-non-cancer, Fibrinogen(+)-non-cancer, Fibrinogen(-)-cancer, Fibrinogen(+)-cancer]. Total RNAs were extracted from the pooled tissue fragments and these samples were analyzed by microarray.
