Genome-wide maps of mRNA state in artesunate and dihydroartemisinin treated mouse neuroblastoma cells with rabies virus infection

To investigate the roles of artesunate (ART) and dihydroartemisinin (DHA) in rabies virus (RABV) infection in mouse neuroblastoma cells, we performed the high-throughput profiling of artesunate and dihydroartemisinin treated mouse neuroblastoma cells combined with rabies virus infection with the RNA sequencing technology. Mouse neuroblastoma cells from 6 groups treated with dimethyl sulfoxide (DMSO), RABV+DMSO, ART, ART+RABV, DHA, DHA+RABV, respectively. The total mRNA from all the mouse neuroblastoma cells was isolated and RNA sequencing was performed using Illumina HiSeq2500. Bioinformatics analysis including differentially expressed genes, relationship analysis of samples, GO enrichment analysis, et al. was performed based on the interested compared groups: DMSO vs RABV+DMSO, DMSO vs ART, DMSO vs DHA, DMSO vs RABV+DMSO vs ART vs ART+RABV, DMSO vs RABV+DMSO vs DHA vs DHA+RABV. We find total 22322 host genes expression with a fragment per kilobase of transcript per million mapped reads (FPKM) value. RNAs differential expression analysis was performed by DESeq2 software between different groups. The genes/transcripts with the parameter of false discovery rate (FDR) below 0.05 were considered differentially expressed genes/transcripts. This study provides a comprehensive profiling of mRNA towards the differential treated cells with ART or DHA in RABV infection in mouse neuroblastoma cells.
