Transcriptional profiling of circulating gamma-delta T cells in psoriasis vulgaris

Gamma-delta T cells are emerging as influential yet understudied contributors to psoriasis vulgaris (PV), a chronic T cell-mediated dermatosis with substantial systemic footprint. Although altered distribution and activation of gamma-delta T cells in blood and skin have been reported, their intrinsic transcriptional changes in PV remain poorly understood. In this study, we performed targeted immunotranscriptomic profiling of FACS-sorted peripheral blood CD3+gdTCR+ T cells from 12 well-characterized PV patients and 11 age- and sex-matched healthy controls. Using the AmpliSeq Immune Response Panel (395 genes) and Illumina MiniSeq sequencing, we identified 36 differentially expressed genes, with 30 showing upregulation in PV. The transcriptional profile indicated an activated effector phenotype with cytotoxic and tissue-homing signatures in circulating gamma-delta T cells, most prominent in a subset of patients. These findings provide novel insights into activation-associated changes in the peripheral gamma-delta T cell compartment in PV.
