Psoriasis is a chronic autoimmune recurrent inflammatory disease with significant erythema and scales as the main clinical features that occur in the trunk, limbs, and scalp, causing serious physical and mental burden on patients. It is widely accepted that the immune system is involved in psoriasis pathogenesis. Adaptive immunity plays a significant role in psoriasis, and Th1 and Th17 inflammatory responses are the key mechanisms involved in psoriasis. Recent studies confirm that innate immunity is necessary for the pathogenesis of psoriasis, such as mast cells and neutrophil.In our study, C57BL/6 Mrgprb2 flox/flox (Mrgprb2fl/fl) mice were purchased from the Chengdu Gembio Co.,Ltd. Cpa3-Cre mice were obtained from The Jackson Laboratory, and used for Cpa3Cre/+/Mrgprb2fl/fl mice hybridization. the backs of C57/BL6 wild type (WT) and Cpa3Cre/+/Mrgprb2fl/fl mice (MUT) mice were shaved, and the IMQ-induced psoriasis model was prepared by topically applying 5 mg of Aldara cream (containing 5% IMQ, INova Pharmaceuticals). After treated with IMQ for 5 days, the mice were sacrificed through CO2 inhalation and skin tissues were collected and RNA-seq was executed.
