Hypothyroidism (HT), defined by inadequate low production of thyroid hormone by the thyroid gland, is a common disorder, particularly in women of childbearing age. HT during pregnancy has many potential etiologies, in particular, IQ deficits in newborns. Recent studies have demonstrated that the composition of the maternal gut microbiome contributes to maternal and fetal long-term health outcomes. However, the relationships between thyroid functions and the maternal microbiome during the pregnancy have not been fully assessed. Therefore, in this proof-of-principle study, we tested whether HT during pregnancy is associated with changes to the maternal oral and gut microbial diversity and composition. In this study, the oral and gut microbial composition was surveyed using 16S rRNA sequencing on 40 HT cases and 96 non-HT controls. We further compared the overall microbial diversity, the differential taxa features, and elucidated the co-occurrence and co-exclusion microbiome network associated with HT.
