snRNA-seq of human fetal lung, adult lung, and fetal-derived lung organoids across the proximal-distal axis

To characterize the complement of cell types and niches with positional specificity, we performed micro-dissections to isolate four different regions of the human lung; the trachea, large cartilaginous airways (bronchi), small non-cartilaginous airways, and distal/peripheral lung during the late pseudoglandular stage. Primary organoids were also established from these four fetal regions. We performed analogous dissection on adult lung donated from Gift of Life from healthy adult trachea, bronchi, and bronchioles. We profiled gene expression using single-nucleus RNA-sequencing (10X Genomics) on these samples (fetal and adult primary tissue, fetal airway organoids) which are presented here.
