The caudal-related homeobox transcription factor CDX2 is ectopically expressed in the majority of patients with acute myeloid leukemia (AML). We generated an inducible transgenic mouse model whereby Cdx2 was specifically activated in HSCs. Cdx2 mice developed myelodysplastic syndrome (MDS) with progression to acute leukemia associated with stepwise acquisition of additional driver mutations. To gain insight into the mechanism underlying Cdx2-mediated transformation of HSCs, we examined RNA-Seq gene expression on Scl-CreERT, Scl-CreERTxCdx2, Scl-CreERT x FLT3 ITD/+ and Scl-CreERT x Cdx2 x FLT3 ITD/+ 4 weeks after tamoxifen induced Cre-Recombinase excision leading to ectopic Cdx2 expression
