DNA extraction optimisation for minute land snails of the genus Vertigo O.F. Müller, 1773 (Gastropoda: Vertiginidae): a comparative evaluation of six methods including a non-destructive shell-preserving protocol

This dataset contains summary data supporting the principal numerical tables and figures in the revised manuscript "DNA extraction optimisation for minute land snails of the genus Vertigo O.F. Müller, 1773 (Gastropoda: Vertiginidae): a comparative evaluation of six methods including a non-destructive shell-preserving protocol" (Ecology and Evolution, manuscript ECE-2026-05-01399). Six DNA extraction methods were compared using Vertigo antivertigo preserved in 96% ethanol (n = 10 per method). The workbook includes method characteristics; QuantiFluor dsDNA concentration and NanoDrop purity summaries; formula-derived total-DNA approximations based on nominal elution volumes; PCR amplification outcomes across COI, COI mini, ITS1 and ITS2; Bioanalyzer 200–1,000 bp fragment summaries; cost estimates supporting revised Figure 8; the aggregate collection-period allocation reported as Table S3; and GenBank accession metadata for PZ227160, PZ227161, PZ227162, PZ236467 and PZ235182. Specimens were collected under permit No. 600000-S-55 (Lithuanian Environmental Protection Agency, 2020-06-08). Version 3 corrects the journal metadata and adds the revised total-yield, allocation and accession information. Specimen-level identifiers and exact collection years were not retained and are not deposited.
