C57BL/6J and DBA/2J mice were delivered (-)-Nicotine hydrogen tartrate [(-)-1-methyl-2-(3- pyridyl) pyrrolidine (þ)-bitartrate] in 0.9% sodium chloride solution by osmotic minipump for 14 days. Mice in the chronic cohort were delivered nicotine up to moment of sacrifice. Mice in the withdrawal cohort had the minipumps surgically removed 24 hours before sacrifice. Nucleus accumbens tissue was dissected and snap frozen in liquid nitrogen. RNA was then extracted and hybridized to Affymetrix Mouse Transcriptome 1.0 arrays for transcriptome quantification. Affymetrix MTA 1.0 arrays were used to determine the transcriptomic effect of chronic nicotine delivered by minipump, or nicotine withdrawal, in the nucleus accumbens of C57BL/6J or DBA/2J mice.
