<b>Neurological deficit assessment</b>Neurological scores of rats in different groups were assessed at 12 h post HS according to the modified Neurological Severity Score (mNSS) system (<i>Alam, Ur Rehman & Ok Kim,</i> <i>2019</i>). Briefly, the scoring system included four parts: the motor test (scores of 0–6), sensory test (scores of 1–2), balance test (scores of 0–6), and reflex measurement (scores of 1–4). Then, the neurological function was graded on scale ranging from 0 to 18 by a blinded investigator, in which 0 point is normal score and a score of 18 represents maximal deficit.<b>Sample collection and brain edema assessment</b>The cerebral edema formation was determined in using measurement of brain water content at 12 h after HS according to previously published procedure with some modification (<i>Gao et al.,</i> <i>2017</i>). For this purpose, the rats were decapitated under general anesthesia, and the brain were rapidly removed and placed on a silver paper. Subsequently, we immediately weighted the brain sample to get the wet weight and dried in an incubator maintained at 80°C for 72 hours to obtain dry weight. The brain water content was defined as the ratio of (wet weight-dry weight)/wet weight×100%.<b>Morris water maze test</b>The Panlab Smart 3.0 system was employed for the Morris water maze (MWM) test at 12 h after HS. The MWM consisted of the spatial learning ability task and spatial memory task of rats. The apparatus is a black circular barrel (120 cm diameter, 50 cm height) with opaque water. The area of water surface was conceptually divided into four quadrants of equivalent size and a circular platform was hidden 1 cm below the water surface in the middle of quadrant III. The spatial learning ability task was repeated 4 trials a day for 5 consecutive days. In all trials, rats were placed into the water from different quadrants and allowed to swim 60 s to explore the platform. Rat escape latency (EL) time required and the distance traveled to find the hidden platform were chosen as the measurement index. If the rat found and landed on the submerged platform in 60 s, the swimming time will be the EL; if the rat failed to find the platform within 60 s, the rat was guided to the platform to stay there for 15 s and the EL of 60 s was assigned for the analysis. On the 6th day, the platform was removed for a spatial memory task. The rats swim freely in the pool for 60 s starting from the same location, and the percentage of the time in the target quadrant and the number of the rats crossed the position where the platform had previously been located were recorded.
