HMGN3 promote metastasis through co-suppress epithelial genes expression with SNAI2 in Cholangiocarcinoma

ChiP sequence data. HMGN3 is a member in nucleosome binding HMGN family that affect chromatin structure and regulate genes expression. In this study we demonstrate the expression and functional roles of HMGN3 in cholangiocarcinoma (CCA). Human CCA tissues shown significant higher HMGN3 expression than normal bile duct. High HMGN3 was related with metastatic status of CCA patients. HMGN3 protein was higher in CCA cell lines than immortalized normal cholangiocyte. Suppression of HMGN3 expression significantly reduced colony formation, migration and invasion of CCA cell lines. HMGN3 significantly related with EMT process and shown enriched in SNAI2 binding sequence in CCA cells. HMGN3 had protein-protein interaction with SNAI2 and either HMGN3 or SNAI2 suppression induced epithelial genes expression. Suggesting that HMG3 and SNAI2 are co-repress epithelial genes expression to promote CCA cell migration. HMGN3 may be the novel alternative targets for CCA treatment.
