Leishmania guyanensis strain:M4147 Genome sequencing

Leishmania guyanensis M4147 is a WHO reference strain (MHOM/BR/75/M4147). This line was obtained from Jean Patterson (Texas Biomedical Research Institute, San Antonio, Texas) and a clonal line (LUC3) was engineered to express high levels of firefly luciferase through transfection of SwaI-digested pIR2SAT-LUC(b) and integration into the SSU rRNA locus as described (Lye et al PLoS Pathogens 2010). These parasites express high levels of luciferase and show virulence in susceptible mouse models comparable to that of the parent M4147 line (Ives et al Science 2011). Lgy M4147 contains the dsRNA totivirus LRV1 (Widmer et al PNAS 1989) and this was confirmed for LgyM4147-LUC3 (Lye et al. reference above). An updated sequence for LRV1-LgyM4147 (Brettman et al PNAS 2016) was deposited in GenBank (KX808487).
