Dataset of cell viability and secreted analytes tumour cell lines treated with low pH and conditioned medium

Extracellular acidity has a profound effect on tumour cell survival and growth, tumour development, and disease progression. Breast and oesophageal squamous cell carcinoma cell lines (MCF-7 and WHCO6, respectively) were treated with medium of decreasing pH, and culture supernatant was removed after 24 hours. Cytokines, chemokines, and growth factors were detected by Luminex. Various levels of apoptosis was induced in acid-exposed cells. This conditioned supernatant was applied to fresh cell cultures for 24 hours and the same proteins were again measured by Luminex. Exposure to conditioned medium had paradoxical effects on both cell lines, with proliferation and survival increasing WHCO6 cells, whereas MCF-7 cells underwent apoptosis. The proteins secreted by these cell lines highlight the differences in survival strategies such as avoiding immune destruction and promoting tumour cell growth. Protein concentrations were determined (pg/ml) in duplicate. One- and two-way ANOVAs followed by post-hoc Dunnett's tests were used for multiple comparisons. Statistical analysis was performed using GraphPad Prism for Windows version 9.4 or 9.5 (California, USA).
