资源简介:
The goal of this experiment is to determine the effect of ATRX depletion on telomeric chromatin compaction. We adapted the ATAC-seq to quantify telomere chromatin accessibility. By scoring the ratio of sequence reads that contained 3 - 12 tandem telomeric repeats over the total number of reads in wild-type and ATRX deplted cells, our analysis revealed that CRISPR/Cas9-mediated ATRX depletion induces a progressive telomere de-compaction. Overall design: ATACseq profiles of wild-type control and ATRX-depleted human IMR90 or BJ fibroblast cells were generated by deep sequencing, in duplicate, using Illumina NEXTSeq 500. The sequence reads that contained 3 - 12 tandem telomeric repeats in wild-type and respective ATRX deplted cells were scored. The ratio of telomere reads over the total number of reads were used to quantify the telomereic nucleosomal compaction. ----------------------------------------------- Authors state "The processed data is what we used for generating the figure of our manuscript. It is telomeric repeat sequence that cannot be processed in the form or wig, bedgraphs or peak files. Only useful data files in fact fastq and excel files we provided."
