Mass cytometry (CyTOF) dataset of early signaling in primary human CD8+ T cells upon target-cell recognition

This dataset contains the single-cell mass cytometry data underlying Figure 6 and Supplementary Figures S10 and S11 of Kotowski et al. 2026 (Nature Communications). The experiment was performed to measure early phosphorylation events during T-cell activation. Experiments were performed using co-cultures of primary human CD8+ T cells with cancer cells presenting T-cell receptor ligand (ImmTAC) and adhesion molecule (ICAM-1) at different surface densities. PMA/ionomycin and pervanadate, which strongly stimulate T cells, were used as positive controls. Unstimulated T cells were used as a negative control. Measurements were taken at four timepoints (1, 5, 15 and 30 minutes) from initial interaction between the T cells and cancer cells or stimulant treatment. The panel contained 43 antibodies; see Supplementary Table S8 of the article for details. Sample_Annotations_CyTOF.txt describes each .fcs file in new_fcs_files.zip, and panel.txt specifies the markers used in downstream analysis.
